    Öncelikle hormon düzeylerine bazal zamanda bakılmalı. Yani adet kanamasının başlamasından itibaren 3 gün içinde. Bunun dışında bakılanlar değerlendirmeye alınmaz. Normalde FSH, LH'ın 2 katı veya üzerinde olmalıdır. Eğer bu oran tersine dönerse, yani LH, FSH'ın 2 katı olursa bu, polikisitik over hastalığı için bir hormonal belirteçtir. Diğer yandan her hormonal belirteç ile polikistik over hastalığı tanısı konamayacağı gibi her polikistik over hastalığında hormonal belirteç olmak zorunda da değildir. Polikistik over hastalığı tanısı için hormonal dışında klinik ve sonografik belirteçler de dikkate alınmalıdır. Bunlar doğrultusunda yumurtlamanız olup olmadığı değerlendirilebilir.
